Research Overview

P21 (more formally P021 in the primary literature) is a synthetic peptide derived from the biologically active region of human ciliary neurotrophic factor (CNTF) - it is NOT a fragment of Cerebrolysin, despite common claims. Preclinical studies in 3xTg-AD and traumatic brain injury models report enhanced neurogenesis, synaptic plasticity, and memory, and reduced tau pathology. Evidence is preclinical only; no human clinical trials establish efficacy, and P21 is not an approved drug. Naming note: 'P21' is also used for the unrelated cell-cycle inhibitor p21 (CDKN1A); these are distinct. Exact peptide length/sequence is not uniformly reported across secondary sources and is not asserted here.
