Mechanism / Biological Target

Acts at the pituitary GHRH receptor to stimulate pulsatile growth hormone release; the tetrasubstituted sequence resists DPP-IV proteolysis. Lacking DAC, it is not albumin-bound and is cleared rapidly.

Research Overview

CJC-1295 No DAC (also called modified GRF 1-29) is a tetrasubstituted analog of growth hormone-releasing hormone(1-29) WITHOUT the C-terminal maleimide 'Drug Affinity Complex' (DAC) that enables albumin binding. Without DAC it does NOT covalently bind albumin and has a short plasma half-life (~30 minutes), unlike CJC-1295 with DAC (half-life ~8 days), which is the form studied in published pharmacokinetic and pharmacodynamic trials. Do not conflate the two compounds: the published human GH/IGF-1 data concern the DAC-containing form. Human evidence specific to the 'No DAC' variant is very limited and it is not an approved drug.
